In vitro anticancer activity of native and modified black rice flour against colon cancer cell line (HCT116)

Abstract:

Pigmented rice such as black rice is known for its health promoting effects and functional activities, owing to the high contents of flavonoids, phenolic compounds, anthocyanins and dietary fibre. Being as a functional food source, black rice provides potential benefit in the prevention of cancer. In this study the modification techniques applied for the preparation of sample were enzymatic modification and heat moisture treatment, with the objective to evaluate cytotoxicity of native and dual modified black rice flour against human colorectal adenocarcinoma cell line (HCT116) and mouse embryo fibroblast cell line (3T3-L1) by using the MTT assay. The data were explored using descriptive and inferential statistical analysis by using the Microsoft Excel Spread sheet 2013 and Scientific Package for Social Sciences version 20.0 software. In this study, the IC50 of NBRF and DMBRF were 255.78 and 340.85 µg/mL, respectively. The result confirms that the NBRF having significant cytotoxic and anti-cancer potential against human colon cancer cells. Also the IC50 values of NBRF and DMBRF on 3T3-L1 cell line were 345.96 and 1106.94 µg/mL, respectively. Therefore, it was proved on normal cell line, the NBRF had weak cytotoxicity and DMBRF is a non-toxic. Black rice contains high levels of phytochemicals and has potential role in the prevention of colon cancer. Black rice as a cancer chemo preventive dietary agent represents a unique approach to evaluate effective whole food compared to the individual phytochemical;
