Analysis of high molecular weight glutenin subunits in a core collection of drought resistant wheat varieties

Abstract:

In order to investigate the combinations and the variations of high molecular weight glutenin subunits, a core collection of drought resistance wheat varieties including 203 wheat cultivars was used in this study. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) results revealed that forty-two types of high molecular weight glutenin wheat subunit combinations were detected. Of the materials tested, 15 subunit combinations had the quality score of over 8.0 point, which accounts for 41.9% of all the materials. Three Glu-A1 variation types were identified, of which subunit one had the highest frequency (up to 58.13%). There were 10 variation types at the Glu-B1 locus, among which subunits 7+8 had a frequency of 48.78%. There were five variation types at the Glu-D1 locus and subunits 2+12 had the highest frequency (up to 45.54%). Compared with Glu-Al and Glu-Dl, Glu-B1 had the most abundant genetic diversity, with a Hannon-Wiener index (H') value of 0.799. Of the HMW-GS combinations detected, the subunit combination type “1, 7+8, 5+10” represented the highest proportion (up to 12.18%).
