Nutrition improvement and pesticide reduction in Napier grass ensiling process with Lactobacillus plantarum and laccase from Ganoderma lucidum

Abstract:

Screening of laccase producing strain for used as microbial inoculum to improve the nutritional values and pesticide reduction of Napier grass silage for ruminant feed, the white rot fungus Ganoderma lucidum was characterized through laccase production. When G. lucidum was incubated in liquid medium at 30°C for 15 d, the value of laccase was achieved at 50.59±7.26 mU/ml. In this study, the optimal condition for silage production was Napier grass at initial moisture 70%, initial pH 6.0, and organochlorine pesticide mixtures and ensiled at 30°C for a period of 21 d. A completely randomized design (CRD) was conducted in which the following 0.2% of three four inoculations were added including LAB inoculum (control: T1), crude laccase (T2), and mixed inoculum in 1:1 ratio between crude laccase and LAB (T3). The results showed that Napier grass ensiled with the addition of both crude laccase and LAB had higher protein content than the control (p<0.05). The analysis of volatile fatty acids from the silage showed that lactic acid was significantly increased in a mixing inoculum. In addition, the laccase was subsequently evaluated for its degradative ability towards several types of organochlorine pesticide mixture. Complete degradation of heptachlor, aldrin, dieldrin, p,p’-DDE, endosulfan, chlordane, and endrin was observed, whereas total BHC, BHC derivatives lindane p,p’-TDE (DDD) p,p’-DDT and, endosulfan were degraded at level greater than between 80 to 90% under these condition of silage processing with the presence of laccase with LAB.
