Abstract:

Twenty Trichoderma strains were isolated from agriculture field soils from Senegal. All the strains were tested for protease activity. Among them, 14 strains showed the alkaline protease activity. Of the 14 strains, maximum protease production was recorded by TG4 and this strain was selected for further studies. The microorganism used in the present study was Trichoderma sp. TG4 was isolated from the soils of agricultural area in Senegal and the strain was identified on the basis of 18S rDNA sequencing analysis. Optimization of fermentation process parameters such as, temperature, pH, incubation period and substrate utilization for alkaline protease production was carried out. The maximum production of alkaline protease by Trichoderma TG4 was found at pH 8, temperature 35°C, when glucose and peptone were used as carbon and nitrogen sources, after 120 h of incubation.
