Antioxidant capacity of Elaeagnus angustifolia L. and investigation of eosin y as the fluorescent probe in ORAC method

Abstract:

Elaeagnus angustifolia L. (oleaster) is one of the widely consumed medicinal plant in Turkey. It is used in the treatment of some diseases in folk medicine. Leaf, seed, peel, and mesocarp parts of oleaster were extracted with methanol (100%, v/v) in ultrasonic bath in the present study, and antioxidant capacities of these parts’ extracts of Elaeagnus angustifolia were evaluated by using oxygen radical absorbance capacity assay (ORAC). The leaf part of Elaeagnus angustifolia L. (428 µmol Trolox Equivalent (TE) g-1) possessed stronger antioxidant properties in comparison with other parts. Fluorescein is the most used fluorescent probe in ORAC method. In this assay, eosin y, which is a derivative of fluorescein, was compared with fluorescein as the fluorescent probe. The detection limit (DL) and quantification limit (QL) were 3.65 and 11.06, 7.12 and 21.57 µM TE for ORAC-fluorescein (ORAC-FL) and ORAC-eosin y (ORAC-EY), respectively.
