Effects of different culture media and flower bud size on haploid plant production through anther culture of three Carica species

Abstract:Effects of different culture media and flower bud size on callus induction and regeneration of haploid plants of C. papaya, C. cauliflora and C. goudotiana were investigated. The half strength MS medium with full strength Na-Fe-EDTA supplemented with 2.0 mg/l NAA, 1.0 mg/l BAP and 60 g/l sucrose exhibited the highest percentage of callus in three Carica species and embryogenic callus as well as haploid plant regeneration in C. goudotiana. The anthers of C. cauliflora induced highest percentage of callus followed by C. goudotiana and C. papaya. The callus of three Carica species induced by different culture media were transferred onto growth regulators free medium. The callus derived from anther of C. papaya and C. cauliflora did not produce embryogenic callus nor plant regenerate plantlets while callus derived from anthers of C. goudotiana produced embryogenic callus and regenerated plantlets. The medium size of flower buds of C. papaya (15-18 mm), C. cauliflora (16-22 mm) and C. goudotiana (12-14 mm) showed the best performance in respect of anthers producing calli in all the three Carica species. The chromosomes from root tip of haploid plantlets derived from anthers of C. goudotiana were counted through cytological observation and nine chromosomes (n = 9) were found confirming the haploid nature of the regenerated plantlets.
