Isolation and germination of Ganoderma lucidum basidiospores and effect of H2Oon the germination of spores

Abstract:

Aseptic environment is needed for both germination of spores and making tissue cultures. The spores of bacteria and other pathogenic fungi can be originated from the environment or can be contaminating agent in the inoculated medium, in this situation, the germinated spores cannot be selected as pure as needed. On the other hand the difficulty of germination frequency is dormancy of spores and all these reasons make pure culture to be obligatory.  The demands to the products which have a pharmacological activity and medical properties, have increased. For this reason, the culture of the mushrooms which are nutriceuticals and natural food source, have increased in comparison with past years. An example of these mushrooms is Ganoderma lucidum which has become popular at biotechnological usage. In this study, a weak acid hydrogen peroxide (H2O2) is used for germinating of basidiospores from basidiocarps of medicinal mushroom Ganoderma lucidum at aseptic conditions. In addition to antiseptic activity of H2O2, it may has a stimulation effect on spore germination.
