Changes of beta-galactosidase and alpha-L-arabinofuranosidase activity and gene expression in relation to fruit storability of apple

Abstract:

The roles of beta-galactosidase (β-Gal) and alpha-L-arabinofuranosidase (α-Af) in ‘Fuji’ and ‘Gala’ apple fruit with different storability and their influence by 1-methylcyclopropene (1-MCP) and cold storage (0°C) were assessed. During storage, the activities and gene expression of β-Gal and α-Af were higher and enhanced more rapidly in ‘Gala’ than  in ‘Fuji’ fruit, which were consistent with a higher respiration rate and a faster decline of firmness in ‘Gala’. 1-MCP treatments significantly inhibited β-Gal activity and gene expression in both types of apple fruit though still showed an increase trend in ‘Gala’ fruit, but for α-Af, the influences by 1-MCP were similar to β-Gal, except no gene expression difference with the control in ‘Fuji’ fruit. Under cold storage, no increase was seen among these behaviors in two cultivars. In addition, β-Gal activity and the related-gene expression were all higher and showed more significant difference between ‘Fuji’ and ‘Gala’ fruit than those of  α-Af. So, it was concluded that β-Gal may play more roles in fruit softening and is more closely related to storability of apple.
