Development of an enzyme-linked immunosorbent assay for detection of ofloxacin residue in environment matrix

Abstract:

Ofloxacin polyclonal antibody was prepared and cross-reactivity to other fluroquinones was observed. Further, a rapid, low-cost ELISA analytical technique was developed, with IC50 at 1.2 ng.mL-1, for the analysis of ofloxacin in the environment matrix including wastewater, manure and sludge. The mean recoveries and coefficients of variation were in a range of 74-83% and 8.5-12.2%, respectively, in wastewater. The mean recoveries and coefficients of variation in pig manure and sludge were from 62% to 78% and from 8.4% to 14.4%, respectively. Concentrations of ofloxacin using ELISA were sometimes greater than measured with LC-MS/MS. It may be due to cross-reactivity to structurally similar compounds or metabolites. Results from this study showed that the ELISA can be used as a preliminary screening tool for later instrumental analysis.
