Rapid and high efficiency regeneration from potato (Solanum tuberosum L.) using thidiazuron as cytokinin source

Abstract:All classic and modern in vitro projects on potato, including site-directed and high through put mutagenesis, somaclonal variation and genetic manipulation of metabolites as well as producing high value materials require a rapid and efficient genotype-independent plant regeneration protocol as an obligation. To reach this objective, we did two individual experiments with thidiazuron in combination with other growth regulators against commonly used protocols containing zeatin using internode and petiole explants of Agria and Marfona cultivars. The comparison of results demonstrated that the regeneration potential in potato still could be dramatically improved using new combinations of growth regulators and appropriate explants type. The superior rate of callus induction and plant regeneration from both explants were recorded as 100% and 141.7 shoots per 6-7 calli, respectively, in the presence of thidiazuron. Whereas, the data obtained from media containing zeatin and absence of TDZ showed only 50% regeneration with the highest rate of 45 shoots per 6-7 calli. The shoots were excised from calli and rooted in the rooting media containing 0.02 mg/l of naphthalene acetic acid, followed by hardening successfully transferred to pods for mini tuberization. All in vitro cycle stages showed phenotypic characteristics as normal plants.
