Initiation of in vitro cultures of Lycopersicon peruvianum var. humifusum

Abstract:The aim of this research was to determine the initiation capacity of wild tomato L. peruvianum var. humifusum. Two kinds of in vitro culture were induced – direct shoot regeneration on explants (apical meristem, axillary bud, leaf base) and indirect via callus. In the first variant 12 concentration combinations of IBA or NAA or KIN or BAP were used and in the second one – 13 combinations of auxin NAA with cytokinin BAP. It was found that clonal micropropagation of L. peruvianum var. humifusum is possible on MS medium with 3 or 4 mg·dm-3 IBA whereas embryogenic callus induction on MS medium supplemented with 0.5 mg·dm-3 NAA and 5 mg·dm-3 BAP. After two months 34 embryos and 1 regenerated plant were obtained from 2.205g of embryogenic callus. Clonal micropropagation, in the majority of cases did not produce genetically homogenous clones.
